SV40 Large T Antigen in Mouse Somatic Cell Reprogramming

Abstract:

SV40 Large T Antigen (TAg) is a well studied transforming protein for animal cells. Recently it was reported that, when used in conjunction with transcription factors such as Oct4 and Sox2, TAg could replace Klf4 and c-Myc in somatic cell reprogramming, and the new combinations of TAg+Oct4 or TAg+Oct4+Sox2 significantly improved the efficiency for the generation of human induced pluripotent stem cells (hiPS). Here we aimed to examine whether TAg has similar effects in inducing mouse embryonic fibroblast cells (MEFs) to mouse iPS (miPS). Retroviral infections of MEF with Oct4+TAg or with Oct4+Sox2+TAg were performed. MEFs transfected with these combinations showed higher cell density over the TAg transfection alone, and Oct4+Sox2 transfection controls. The combinations of Tag+Oct4 or Tag+Oct4+Sox2, however, showed only partial reprogramming effects on miPS induction, when judged by the morphology of colony formations. These observations indicate different reprogramming mechanisms between human and mouse somatic cells. Another possible explanation would be that a TAg/transcription factor ratio dependence exists for successful induction of miPS using TAg+Oct4 or TAg+Oct4+Sox2.
